Construction of RNAi expression vector of the plasma membrane Na╋/H╋ antiporter gene in succulent xerophyte Zygophyllum xanthoxylum

Abstract
Total RNA was extracted from roots of succulent xerophyte Zygophyllum xanthoxylum seedlings, and the fragment of the plasma membrane Na+/H+ antiporter gene ZxSOS1 was amplified by RTPCR. Then the RNAi plant expression vector PARS with inverted repeats of ZxSOS1 driven by the promoter CaMV 35S was constructed based on the intermediate vector pHANNIBAL and the plant expression vector PART27 by restriction enzyme digestion and ligation. The work might be a good basis for further investigation of the roles of ZxSOS1 in the transportation of Na+ and the adaption to arid environment in Z.xanthoxylum by RNAi technique.
