Study on germination conditions of hybrid embryo of Caucasian clover white clover

Sixploid Caucasian clover and tetraploid white clover were used as distant hybridization parents to test the germination conditions of hybrid embryo by using tissue culture. The results showed that the highest germination rate（8%）could be obtained under the conditions of Caucasian clover as the female parent, white clove as the male parent, pollinated at 8:00 to 9:00, the embryo was picked after 12 to 13 days from pollination, the hybrid embryo was sterilized by NaClO（10%）for 6.5 to 7 min, cultured on eight improved MS media. For higher frequency propagation, the germination was higher on the above mentioned medium added with 0.25 mg/L of 2,4D and 0.50 mg/L of 6BA.
