ISSR analysis of genetic differentiation of Stipa bungeana populations in the mountain area of southern Ningxia

Abstract
In order to study and explore the mechanism of molecular ecology of Stipa bungeana, intersimple sequence repeat (ISSR) marker was used to analyze the genetic differentiation and population structure of six natural populations of S.bungeana from the mountain area of southern Ningxia. The results showed that fifteen primers generated a total of 244 amplification bands from the six natural populations, of which 239 (97.95%) were polymorphic bands. Neis gene diversity index and Shannon information index were 0.254 4 and 0.396 6, respectively, which indicated that the high genetic diversity was detected at the species level. AMOVA analysis revealed a high level of genetic variation within population (58.06%) and a relative lower of genetic variation among populations (41.94%). There was no significant correlation between genetic distance and geographic distance by Mantel test.
